Hi,
It doesn't matter which buses you use. I tested it with PCI/PCI, PCI/AGP and PCIe-x16/PCIe-x1. I have no doubt it works also with PCIe/PCI mixed.
For instance, with 5 cards (4x PCI Matrox G200 on one card (a 'quad'), plus one AGP nVidia GeForce2MX400) you get to choose where your Desktop goes via the xxx.settings file's 'primary'setting, and where the video goes is determined trhough the node's preferences tab in Mediaprefs.
The PCIe-x16 was a GF6600 (started by the system BIOS, so displaying POST), and a PCIe-x1 Matrox G550.
One combination that didn't work was the Quad G200 (on one card, PCI), combined with a G200 PCI seperately: this wasn't initialized/started by the PC's system BIOS: in other words: the PC didn't even show POST (never started). This non working combi would have worked in BeOS however if that had been started.
If you run this stuff, let me know how it turns out :-)
Have fun..
Rudolf.
